LAMP1（lysosomal associated membrane protein 1）またはCD107a（cluster of differentiation 107a）は、ヒトではLAMP1遺伝子によってコードされているタンパク質である。ヒトのLAMP1遺伝子は13番染色体（英語版）長腕上の13q34領域に位置している。

LAMP1はLAMPファミリー（英語版）に属する糖タンパク質である^[5]。LAMP1はI型膜貫通タンパク質であり^[6]、少なくとも76種類の異なる正常組織細胞種において高〜中レベルで発現している^[7]。LAMP1は主にリソソーム膜に位置し^[8]、セレクチンへ糖鎖リガンドを与える機能を果たしている^[5]。LAMP1はCD8⁺細胞やNK細胞などのリンパ球の脱顆粒（英語版）のマーカーとなることも示されており^[9]、また腫瘍細胞の分化や転移にも関与している可能性がある。

構造

LAMP1は主にリソソーム膜に位置し、高度なN-結合型グリコシル化を受けた内腔側領域と細胞質に露出した短いC末端テールから構成される^[8]。内腔側領域にはヒンジ様構造が含まれており、ヒトのIgAに観察されるものと相同なジスルフィド結合を形成することができる^[8]。LAMP1の他の特徴としては次のような点が挙げられる。

ゴルジ体のトランス槽を移行するための大量のポリラクトラサミノグリカンとシアル酸の付加^[10]

機能

LAMP1とLAMP2は全リソソーム膜タンパク質の50%を占め^[6]、リソソームの完全性やpH、異化の維持の一部を担っていると考えられている^[6]^[11]。LAMP1とLAMP2の発現は連鎖しており、LAMP1遺伝子の欠陥はLAMP2の発現上昇をもたらす^[11]。そのため、これら2種類のタンパク質はin vivoでは同様の共通した機能を有すると考えられている^[6]。一方でこうした性質はLAMP1の正確な機能の理解を困難なものにしており、LAMP1欠損変異体の表現型はLAMP2のアップレギュレーションのために野生型とほとんど差異が見られず^[6]^[11]、またLAMP1/LAMP2二重欠損変異体は胎生致死となる^[11]。

LAMP1は主にリソソーム膜に位置しているが、特定のケースでは細胞膜に発現していることもある^[11]。LAMP1の細胞膜への発現は、リソソームが細胞膜へ融合することによって生じる場合がある^[12]。細胞表面に発現したLAMP1はセレクチンのリガンドとして機能し^[13]^[14]、細胞間接着の媒介を補助する^[15]。したがって、細胞表面におけるLAMP1の発現は、細胞傷害性T細胞、血小板、マクロファージなど、遊走または浸潤機能を有する細胞で観察される^[16]。LAMP1やLAMP2の細胞表面への発現はがん細胞でも観察されることが多い^[16]^[17]。特に結腸がんや悪性黒色腫など転移能の高いがんで観察され^[16]、細胞表面発現と転移能との相関が示されている^[11]。

がんにおける役割

腫瘍細胞の表面へのLAMP1の発現は多くの種類のがんで観察されており、特に膵がん^[18]^[19]、結腸がん^[16]^[17]、悪性黒色腫^[16]^[17]など転移性の高いがんでみられる。LAMP1は細胞接着や遊走の媒介を補助していると考えられており^[15]^[17]^[18]、LAMP1の糖鎖修飾などの構造は腫瘍細胞の分化^[8]^[20]や転移能^[11]との相関がみられる。事実、一部のがん細胞では細胞外マトリックスへの接着はLAMP1やLAMP2とE-セレクチンやガレクチン（英語版）の間の相互作用によって媒介されており、LAMPは細胞接着分子のリガンドとして機能している^[17]。
